method for preparing nicergoline/chemical synthesis of nicergoline/How is nicergoline made?
Preparation of Nicergoline: To a solution of 1-methyl lumilysergol-10-methyl ether in pyridine, 5-bromonicotinyl chloride is used as an acylating agent at room temperature. The mixture is stirred for 1 hour. Water and methanol are added and the resulting mixture is stirred for 1 hour, extracted with chloroform, and washed in sequence with 1% aqueous caustic soda, 5% aqueous sodium bicarbonate solution, and water. The resulting solution is dried over anhydrous sodium sulfate and the solvent is distilled off. By recrystallization of the residue from acetone petroleum ether, nicergoline is obtained, melting at 136° to 138°C.

Forty six patients suffering from vertigo and tinnitus were treated with nicergoline in a dose of 15 mg/day for four or eight weeks. The efficacy of the drug in the reduction of vertigo was marked improvement in 11.1%, marked or moderate improvement in 62.5% and slight to marked improvement in 88.9% at four weeks.

Nicergoline is a drug used for age-dependent cognitive impairment and it protects cultured neurons against β-amyloid toxicity.
nicergoline mechanism of action
Nicergoline acts by inhibiting the postsynaptic alpha(1)-adrenoceptors on vascular smooth muscle. This inhibits the vasoconstrictor effect of circulating and locally released catecholamines (epinephrine and norepinephrine), resulting in peripheral vasodilation.
nicergoline dosing

nicergoline side effects
The side effects of nicergoline are usually limited to nausea, hot flushes, mild gastric upset, hypotension and dizziness. At high drug dosages, bradycardia, increased appetite, agitation, diarrhea and perspiration were reported.
